Abstract
Despite substantial advancements in antiretroviral therapy (ART), a definitive cure for HIV remains elusive due to several persistent challenges, including drug toxicity, the emergence of resistant viral strains, and the existence of latent viral reservoirs that escape therapeutic eradication. Notably, a rare subset of individuals known as elite controllers possess the ability to naturally suppress HIV replication in the absence of therapy, offering a valuable model for uncovering host-mediated mechanisms of viral control. Several host restriction factors, such as SLFN11, CDKN1A, and SAMHD1, have been identified as critical players in inhibiting viral replication through distinct biological pathways. In this study, a novel gene activation approach utilising CRISPRa technology was explored to upregulate these key antiviral host factors.
